Profile
The antigen formulation team(AFT)is part of the Bacterial Vaccines analytical development team. The team acts as subject matter experts in the biophysical characterization of antigens and the formulation and stability of proteins and glycoproteins. Other task within the team are:
  • Characterization of carbohydrate, protein-, or glycoconjugate-based antigens using liquid chromatography (LC);
  • Optimization and improvement of established tests for carbohydrates, proteins, glycoconjugates, and glycoprotein vaccines for in-process controls (IPC) and release testing of drug substances and drug products;
  • Analyzing the quality of drug substances produced by the BacVac process development teams or by CROs;
  • Development and implementation of new and innovative Analytical Technology Platforms;
  • Stability study of (blended) drug substances and drug products;
  • Characterization of adjuvant systems;
  • Biophysical characterization of large molecules;
  • Discovery stage platform formulation development.

As an associatre scientist you are responsible for two major vaccine projects. In your work, you will interact with various Bacterial Vaccines teams and with Janssen vaccine support departments. In addition, you will be expected to perform laboratory work in person (core part of the job) with following documentation. The Bacterial Vaccines department is strengthening its biophysical assay capabilities centered mainly on multidomain proteins and glycoprotein antigens. This position has, therefore, a strong focus on the pipetting skills and characterization using LC.
Your primary responsibilities will be:
  • Know, implement, and improve established formulation protocols.
  • Analyze protein-, or glycoprotein-based antigens (focus methods: UPLC/UHPLC and plate-based assays);
  • Perform and implement formulation and stability experiments according to designed plans with following release testing;
  • Ensure compliance with regulatory requirements, GMP, and safety.


Who we're looking for
For this role we are looking for Bachelors' or Masters' candidates with at least 2 years of experience in the pharmaceutical industry (GLP/GMP). You have knowledge of analytical literature, with emphasis on biophysical characterization using liquid chromatography. Additionally, you have solid experience with liquid chromatography systems. You need to have understanding of the formulation of large molecules such as protein and conjugates. It is a plus if you also have experience with other techniques such as cSDS-PAGE, SEC-MALS, NanoDSF, UV280, DLS. As a person, you can describe yourself as a team player, result oriented and accurate with an analytical mindset.

Department
The Bacterial Vaccines (BacVac) department is working on the discovery and early development of novel vaccines for bacterial infectious diseases. It strives to address unmet medical needs, such as bacteremia (sepsis), meningitis, respiratory tract, and nosocomial infections. The projects at the Bacterial Vaccines department are primarily structured on scientific and technical collaboration with external biotechnology partners (governmental and industrial) that are innovative leaders in their field. This working process includes ‘state of the art’ vaccine discovery, with subsequent in vivo evaluation of these vaccine candidates in animal models for immunogenicity and/or efficacy. Besides, manufacturing processes are optimized and established, and the resulting clinical trial material is evaluated in human clinical studies.
The antigen formulation team (AFT) is part of the Bacterial Vaccines analytical development team. AFT is responsible for:
  • Developing formulations workflows for novel vaccine candidates;
  • Defining key quality attributes and setting specifications for release testing at discovery/early development stage;
  • Evaluating compatibility of antigen formulations with adjuvants;
  • Developing antigen mixing strategies to facilitate compounding of multivalent vaccines ;
  • Preparation of pre-clinical drug product formulations for animal studies.
