For our client, Janssen Vaccines & Prevention, we are looking for an Associate Scientist Analytical Chemistry.

The Analytical Development (AD) team is part of BacVac and provides subject matter expertise in:
  • Liquid chromatography of carbohydrates and (glyco)proteins (SEC, RP, HILIC, IEX);
  • Mass spectrometry (MALDI, LC-MALDI, LC-MS & CE-MS);
  • Capillary electrophoresis (cGE, CE-SDS, cIEF);
  • Biomolecular assays (capillary Western blot, BLI/Octet);
  • Biophysical characterisation of large molecules (NanoDSF, DSC, FTIR, DLS, MALS).
They are also responsible for: 
  • Development and implementation of new and innovative analytical technologies;
  • Leading analytical investigations for research-grade drug substances and drug products;
  • Offering analytical support to BacVac teams with a focus on strain engineering, process development, vaccine formulation, and batch-to-batch comparison.
As part of the BacVac AD team, you will work on 2-3 vaccine projects and interact with various BacVac teams and Janssen Vaccines departments. You are expected to perform laboratory work in person, collaborate with academic partners and help contract research organisations plan and execute their assignments. 
The BacVac AD team is strengthening its analytical capabilities centred on carbohydrate and glycoprotein antigens. This position has, therefore, a strong focus on elucidating and identifying structures of polysaccharides and O-glycans using liquid chromatography (LC).
Your primary responsibilities will be to:
  • Analyse carbohydrate-, protein-, or glycoconjugate-based antigens using ion chromatography with pulsed amperometric detection (IC-PAD or HPAEC-PAD), UPLC-FLD, RP-UPLC & LC-MS;
  • Know, implement, and improve established tests;
  • Collaborate closely with colleague LC and MS experts in the AD team to determine polysaccharide chain length, glycan fingerprinting, number of repeating units, glycosylation sites and occupancy;
  • Develop and perform (automated) sample preparation work flows including desalting, hydrolysis, fluorescent labelling and clean up;
  • Execute LC methods and perform maintenance and troubleshooting of the instruments;
  • Apply state-of-the-art analytical chemistry to ensure a targeted approach to problem-solving and delivering relevant results. Work in a multidisciplinary team of researchers and show a willingness to learn/explore new techniques;
  • Act quickly and provide expertise and solutions for advanced characterisation of large biomolecules and support investigations. Write test procedures and study reports;
  • Ensure compliance with safety, GLP and regulatory requirements;
  • Support LC equipment selection, purchase, and maintenance;
  • Support CROs to plan, execute, document, and review their work;
  • Stay up to date with scientific literature. 

Who we're looking for
The ideal candidate has an M.Sc. in Analytical Chemistry or Pharmaceutical Sciences with 2-5 years of relevant experience, preferably in an industry setting. You are an LC specialist in carbohydrate analysis (glycans, glycoproteins, polysaccharides) using SEC, RP, NP, etc., and you are willing to learn HPAEC-PAD analysis of polysaccharides. 
Skills that are nice to have are:
  • Classical methods of carbohydrate analysis (hydrolysis, hydrazinolysis, methylation, etc.);
  • Mass spectrometry experience including GC-MS and MS/MS, SEC-MALS of polysaccharides, capillary electrophoresis of conjugates;
  • Knowledge of assay qualification and validation (ICH guidelines);
  • Knowledge of analytical literature, with emphasis on carbohydrates and glycopeptides;
  • Hands-on expertise on structural elucidation of bacterial oligosaccharides.

This is a lab-based job where up to 60% of the time is devoted to hands-on experiments. We are only looking for professionals with a strong affinity for hands-on lab-based activities.

The Bacterial Vaccines (BacVac) department is working on the discovery and early development of novel vaccines for bacterial infectious diseases. It strives to address unmet medical needs, such as bacteremia (sepsis), meningitis, respiratory tract, and nosocomial infections. Projects at the Bacterial Vaccines department are primarily structured on a scientific and technical collaboration with external biotechnology partners (governmental and industrial) that are innovative leaders in their field. This working process includes 'state of the art' vaccine discovery, with subsequent in vivo evaluation of these vaccine candidates in animal models for immunogenicity and efficacy. In addition, manufacturing processes are optimised and established, and the resulting clinical trial material is evaluated in human clinical studies.
The Bacterial Vaccines department has well-equipped laboratories and the possibility to acquire additional equipment and laboratory space. Janssen Vaccines’ departments, such as Vaccine Process & Analytical Development (VPAD) and Quality Control (QC), provide expertise to BacVac and its external partners to develop their prototype vaccines.
